Almost every peptide Certificate of Analysis leans on two tests: HPLC and mass spectrometry. They sound interchangeable, and vendors sometimes present them as if either one alone proves quality. They don’t. They answer two different questions, and the difference is exactly where fake or mislabeled product slips through.
HPLC answers: how pure?
High-Performance Liquid Chromatography separates a sample into its components and measures how much of each is present. The output is a chromatogram — a chart of peaks — and a purity percentage, like “98.6%.” A tall, clean single peak with tiny impurity peaks around it is what you want to see. HPLC is the purity test: it tells you the sample is mostly one substance and not a soup of by-products.
But notice what it doesn’t tell you. HPLC measures how clean the sample is, not what the clean part actually is. A vial can be 99% pure… of the wrong molecule.
Mass spec answers: is it the right compound?
Mass spectrometry measures molecular weight. Every peptide has an expected mass based on its amino-acid sequence. The mass-spec result confirms identity by matching the measured weight to the expected weight for the labeled compound. This is the identity test: it’s what proves the pure substance in the vial is genuinely, say, BPC-157 and not a cheaper look-alike or a different peptide entirely.
Why you need both
Put them together and the logic is simple. HPLC without mass spec: “this is very pure something.” Mass spec without HPLC: “the right compound is in here, but we don’t know how much junk is with it.” Only both together give you the full statement a buyer actually needs — this is the right compound, and it’s clean.
